NOVELTY - Immunosensor construction process involves selecting graphite with carbon nanotubes and gold nanoparticles (screen printed DRP 110-GPH-GNP110) as working electrode or other working electrodes, combining nanotechnological materials such as polymer films, graphene, carbon nanotubes and nanoparticles. The anti-PGL1 antibody, anti-LAM or other peptide/mimetic antibody are immobilized. The Rhodamine 6G electrochemical indicator is incorporated, and biological fluid samples are used. The oxidation peaks of Rhodamine 6G identified with potential between 0.30 and 0.95V is identified. USE - Immunosensor construction process for indirect electrochemical detection of mycobacteria, as biological samples saliva, blood, urine, tear, breast milk, semen, sweat and tissue samples, and used in the detection of proteins, which do not have oxidizable amino acids, such as antigen, antibodies, peptides, enzymes or any other proteins or fragments of biological or synthetic proteins, used to obtain a lyophilized Kit containing magnetic beads coupled to protein G and/or antibodies (such as PGL1/LAM) and/or other antigens specific for mycobacteria, followed by consecutive steps or in alternating orders of BSA, rhodamine 6G, adding biological samples in order to carry out the suspension, after this procedure, magnet is added to attract the magnetic beads thus occurring the magnetic capture on the surface of the electrode, in sequence the electrochemical responses are evaluated allowing the diagnostic differentiation (all claimed). ADVANTAGE - The immunosensor construction process has rapid diagnosis of these pathogens, applicability, sensitivity, selectivity and low cost.
