ADVANTAGE - The method: solves the problems of thick ice layer, severe orientation advantage and protein denaturation caused by gas-liquid interface when preparing cryo-electron microscope samples. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for preparing the electron microscope carrier net using hydrophobic protein film as supporting film, comprising dripping the hydrophobic protein solution onto a substrate with hydrophobic properties, leaving it to stand still, forming hydrophobic protein film on the surface of the drop, taking the electron microscope carrier net, placing it on the surface of the droplet so that the electron microscope carrier net is in contact with the hydrophobic protein film, incubating until the hydrophobic protein film is spread on the electron microscope carrier net, removing the excess liquid on the electron microscope grid covered with hydrophobic protein film, rinsing the electron microscope carrier net covered with hydrophobic protein film with pure water dropwise, and naturally drying the rinsed electron microscope carrier net to obtain an electron microscope carrier net.
