NOVELTY - Preparation of three-dimensional graphene marker involves immersing glassy carbon electrode in cyclodextrin-functionalized graphene solution, processing, drying, washing with ultrapure water, drying, then immersing resultant electrode in bovine serum albumin solution, drying, washing with ultrapure water, processing in refrigerator at 4 degrees C, immersing resultant electrode in antigen solution, drying, washing with ultrapure water, immersing resultant electrode in platinum/palladium-copper-based three-dimensional graphene-labeled antibody solution and processing. USE - Preparation of three-dimensional graphene marker used as tumor marker chosen from squamous carcinoma antigen, carcinoembryonic antigen, alpha -fetoprotein, breast cancer susceptibility gene, ovary cancer carbohydrate antigen and prostate specific antigen in sandwich-type electrochemical immunosensor (all claimed). ADVANTAGE - The three-dimensional graphene marker has excellent sensitivity, and enables early diagnosis of tumor. DETAILED DESCRIPTION - Preparation of three-dimensional graphene marker involves polishing 4-mm diameter glassy carbon electrode using alumina polishing powder, cleaning using ultra-pure water, immersing electrode in 6 mu L 0.5-1.5 mg.mL-1 cyclodextrin-functionalized graphene solution, processing, drying at room temperature, washing with ultrapure water, drying, then immersing resultant electrode in 3 mu L 1 %mass bovine serum albumin solution, drying, washing with ultrapure water, processing in refrigerator at 4 degrees C, immersing resultant electrode in 6 mu L 0.0001-30 ng/mL antigen solution, drying, washing with ultrapure water, processing in a refrigerator at 4 degrees C, immersing resultant electrode in 6 mu L platinum/palladium-copper-based three-dimensional graphene-labeled antibody solution, processing and placing in refrigerator at 4 degrees C.
